A column chromatograph works by separating compounds of a mixture according to their molecular weights, amphiphilicity, and solubility. The mixture is added to a column filled with an absorbent material that holds the mixture in place while a liquid solvent is passed through the column. As the solvent passes through, different compounds are adsorbed onto and eluted off the column depending on their physical and chemical characteristics. Compunds with higher hydrophobicities, or those that contain non-polar or hydrocarbon-based atoms, will tend to adsorb more onto the column while compounds that contain more polar or hydrophilic atoms typically have a lesser affinity for the absorbent material and are more quickly eluted off the column. By the end of the separation, the compounds of the mixture will have been separated according to their molecular weight and chemical characteristics.
