How do you test the effect of RpoS on oxyR expression? I cannot find information on this topic anywhere... please help me out! Thanks you!

Additionally, the RpoS mutant strain should be treated with a chemical or drug that increases RpoS expression, and the quantity of oxyR transcripts should be measured again. By comparing the quantity of oxyR transcripts in both the mutant and the RpoS-treated strain with those in the wild-type strain, it is possible to determine the effect of RpoS on oxyR expression. Alternatively, the oxyR promoter could be cloned and inserted into a reporter plasmid, and this plasmid can be transformed into either the RpoS mutant or the wild-type strain. The quantity of the reporter protein that is produced can then be measured, and this indicates the level of oxyR expression in each strain.
